The Role of Aromatic Interactions in Biomolecular Recognition: Contributions to Affinity and Specificity
نویسنده
چکیده
Using beta-hairpin model systems, we have investigated the role of aromatic interactions in providing selectivity to protein folding and protein-protein interactions. In particular, we have explored the role of cation-p and amide-p interactions and their role in controlling proteinprotein interactions with post-translationally modified proteins. Specific modifications that have been studied are Lysine methylation and acylation and Arginine methylation. The molecular recognition of these modified amino acids has particular relevance to understanding the “histone code”, which has been proposed to control chromatin structure and gene expression.

Optimizing Scoring Function of Protein-Nucleic Acid Interactions with Both Affinity and Specificity
Protein-nucleic acid (protein-DNA and protein-RNA) recognition is fundamental to the regulation of gene expression. Determination of the structures of the protein-nucleic acid recognition and insight into their interactions at molecular level are vital to understanding the regulation function. Recently, quantitative computational approach has been becoming an alternative of experimental techniq...
متن کاملSpecificity and Affinity Quantification of Flexible Recognition from Underlying Energy Landscape Topography
Flexibility in biomolecular recognition is essential and critical for many cellular activities. Flexible recognition often leads to moderate affinity but high specificity, in contradiction with the conventional wisdom that high affinity and high specificity are coupled. Furthermore, quantitative understanding of the role of flexibility in biomolecular recognition is still challenging. Here, we ...
متن کاملDynamics Govern Specificity of a Protein-Protein Interface: Substrate Recognition by Thrombin
Biomolecular recognition is crucial in cellular signal transduction. Signaling is mediated through molecular interactions at protein-protein interfaces. Still, specificity and promiscuity of protein-protein interfaces cannot be explained using simplistic static binding models. Our study rationalizes specificity of the prototypic protein-protein interface between thrombin and its peptide substra...
متن کاملCation-pi bonding and amino-aromatic interactions in the biomolecular recognition of substituted ammonium ligands.
Cation-pi bonds and amino-aromatic interactions are known to be important contributors to protein architecture and stability, and their role in ligand-protein interactions has also been reported. Many biologically active amines contain substituted ammonium moieties, and cation-pi bonding and amino-aromatic interactions often enable these molecules to associate with proteins. The role of organic...
متن کامل1,1'-Binaphthyl-based imidazolium chemosensors for highly selective recognition of tryptophan in aqueous solutions.
A type of 1,1'-binaphthyl-based imidazolium chemosensor module has been synthesized for the highly selective recognition of tryptophan (Trp) among the eleven alpha-amino acids investigated in aqueous solutions via synergistic effects of multiple hydrogen bonding and electrostatic interactions. These results have demonstrated that the C-2 hydrogen atom of the imidazolium ring plays a key role as...
